A new high‐resolution structure of a pain‐sensing ion channel, TRPA1, provides a molecular scaffold to understand channel function. Unexpected structural features include a TRP‐domain helix similar to TRPV1, a novel ligand‐binding site, and an unusual C‐terminal coiled coil stabilized by inositol hexakisphosphate (IP6). TRP‐domain helices, which structurally act as a nexus for communication between the channel gates and its other domains, may thus be a feature conserved across the entire TRP family and, possibly, other allosterically‐gated channels. Similarly, the TRPA1 antagonist‐binding (...) site could also represent a druggable location in other ion channels. Combined with known TRPA1 functional properties, the structural role for IP6 leads us to propose that polyphosphate unbinding could act as a molecular kill switch for TRPA1 inactivation. Finally, although packing of the TRPA1 membrane‐proximal region hints at a mechanism for electrophile sensing, the details of how TRPA1 responds to noxious reactive electrophiles and temperature await future studies.Also watch the Video Abstract. An interdisciplinary theoretical framework is proposed for analysing justice in global working conditions. In addition to gender and race as popular criteria to identify disadvantaged groups in organizations, in multinational corporations (MNCs) local employees (i.e. host country nationals (HCNs) working in foreign subsidiaries) deserve special attention. Their working conditions are often substantially worse than those of expatriates (i.e. parent country nationals temporarily assigned to a foreign subsidiary). Although a number of reasons have been put forward to justify such inequalities—usually with (...) efficiency goals in mind—recent studies have used equity theory to question the extent to which they are perceived as fair by HCNs. However, since perceptual equity theory has limitations, this study develops an alternative and non-perceptual framework for analysing such inequalities. Employment discrimination theory and elements of Rawls’s ‘Theory of Justice’ are the theoretical pillars of this framework. This article discusses the advantages of this approach for MNCs and identifies some expatriation practices that are fair according to our non-perceptual justice standards, whilst also reasonably (if not highly) efficient. Psychologists of my generation will recognize the implicit reference in my title immediately: to Kurt Lewin's classic paper that introduced most of us to the excitement of his ideas when we read it as the initial chapter of A Dynamic Theory of Personality . When Lewin wrote about "The Conflict Between Aristotelian and Galilean Modes of Thought in Contemporary Psychology" over a half a century ago, it was indeed a breath of fresh air. Along with a very few other books (...) and papers, it stands out saliently in my memory as having had a large part in forming my enduring perspective in psychology. It surely played a similar role for many others, by no means just Lewinians. We cannot readily recall its content since we've absorbed it, and built it into the fabric of our thought. I turn back to Lewin's essay because it represents the psysicalist tradition in psychological theory at its best, free of many faults that tainted the behavioristic expressions of positivism. All the same, the hermeneutic and contextualist critique of positivism should leave us dissatisfied with Lewin's version of a Galilean strategy for psychology. In one respect, thus, I am using this occasion for my own "me-too" endorsement of post-positivist theorizing. My second concern here is to focus attention on the need for a culturally and historically contextualized approach in personality theory. Finally, this occasion lets me talk some sense about post-positivist perspectives in psychology. As usual, the conceptual innovators have not been very reasonable. If a contextualized psychology of persons is to be advanced, we need a more plausible version of contextualism than is being argued by the leading polemicists. This is also an effort, therefore, to domesticate a contextualized approach, to accommodate it to its prospective role of defining a new mainstream of theorizing at the softer, more human end of the psychological spectrum. 2012 APA, all rights reserved). Robbins, Francis, and Rutledge documented the personality profile of Church of England clergymen and clergywomen prior to the ordination of the first women to the priesthood in 1994, drawing on Eysenck's three-dimensional model of personality. They found that the personality profiles of clergymen and clergywomen were indistinguishable. The present paper reports a comparable study conducted in 2004 among 182 clergywomen and 540 clergymen serving in similar parochial posts in order to examine whether the ordination of women to the priesthood had (...) impacted the overall personality profile of Anglican clergy. The data suggest that little change had taken place between the two cohorts of clergy studied. Once again clergywomen and clergymen appeared to be formed in the same image. (shrink)

The co-occurrence of stress-induced posttraumatic stress disorder (PTSD) and obesity is common, particularly among military personnel but the link between these conditions is unclear. Individuals with comorbid PTSD and obesity manifest other physical and psychological problems, which significantly diminish their quality of life. Current understanding of the pathways connecting stress to PTSD and obesity is focused largely on behavioral mediators alone with little consideration of the biological regulatory mechanisms that underlie their co-occurrence. In this work, we leverage prior knowledge to (...) systematically highlight such bio-behavioral mechanisms and inform on the design of confirmatory pilot studies. We use natural language processing (NLP) to extract documented regulatory interactions involved in the metabolic response to stress and its impact on obesity and PTSD from over 8 million peer-reviewed papers. The resulting network describes the propagation of stress to PTSD and obesity through 34 metabolic mediators using 302 documented regulatory interactions supported by over 10,000 citations. Stress jointly affected both conditions through 21 distinct pathways involving only two intermediate metabolic mediators out of a total of 76 available paths through this network. Moreover, oxytocin (OXT), Neuropeptide-Y (NPY), and cortisol supported an almost direct propagation of stress to PTSD and obesity with different net effects. Although stress upregulated both NPY and cortisol, the downstream effects of both markers are reported to relieve PTSD severity but exacerbate obesity. The stress-mediated release of oxytocin, however, was found to concurrently downregulate the severity of both conditions. These findings highlight how a network-informed approach that leverages prior knowledge might be used effectively in identifying key mediators like OXT though experimental verification of signal transmission dynamics through each path will be needed to determine the actual likelihood and extent of each marker’s participation. In his ‘Theory of Light and Colours’, presented to the Royal Society in November 1801, Thomas Young defended a mechanical explanation of the coloured fringes observed outside of the shadow of an opaque object – the so-called ‘colours by inflection’ – that was based on the hypothesis of an ethereal density gradient surrounding all material bodies. However, two years later, he publicly rejected that hypothesis, without giving much detail of his reasons. Although Geoffrey Cantor has demonstrated the crucial role of (...) mechanical and astronomical arguments in explaining the withdrawal of this fundamental hypothesis long ago, the purpose of this article is to draw deeper attention to a set of experiments performed by David Brewster on the inflection of light, described in a letter he addressed to the Royal Society in January 1802, but was retained by William Herschel, and finally was neither read before the Society, nor published in its Philosophical Transactions. For the hypothesis that will be evaluated here is that these unpublished experiments of Brewster’s were eventually known to Young through the mediation of Herschel, and eventually played a significant role in Young’s rejection of his ether distribution hypothesis. ABSTRACT The imagination has always been thought to operate primarily in conjunction with the sense of vision, imagined objects and scenes being conjured up before the ‘mind’s eye’. In early nineteenth-century Scotland the natural philosopher David Brewster developed a theory of the imagination that explained its operation through a reversal of the normal processes of visual perception. These ideas were rooted in the mental philosophy of the eighteenth-century Scottish Enlightenment. For Brewster the mind’s eye was also the eye (...) of the body, and images from the memory and imagination were projected onto the retina in the same manner that images were projected onto the screen in a magic lantern show. This theory underpinned his belief that imagination played an essential role in scientific discovery. Brewster believed that this process was as essential to the discoveries of science as it was to the creation of great poetry. The writings of Brewster can tell us a great deal about the connections between science and literature in the early nineteenth century, as well as showing that the philosophy of science in contemporary Britain was far from monolithic. Currently, there is no consensus guideline as to the type or degree of donor steatosis considered acceptable for liver transplantation, and little is known about local practices in this area. The aim of this survey (...) was to evaluate current clinical practices amongst liver transplant surgeons in Australia and New Zealand in the evaluation and use of steatotic donor livers in LT.Methods: An anonymous online twelve-question survey was emailed to all practicing LT surgeons in ANZ in January 2010.Results: The response rate was 83%. Estimated prevalence of steatosis in donor livers was between 40% and 60%. In determining suitability for LT, 90% of respondents reported rejecting organs with "severe" steatosis based on visual and palpation grounds alone. A total of 68% sought further histological assessment if the donor liver looked bad and there were risk factors for steatosis. The majority of respondents performed only one biopsy of the liver, using hematoxylin and eosin staining for fat assessment. There was wide variation in the upper limit of steatosis considered to be acceptable for LT. A total of 21% of respondents still considered microvesicular steatosis a risk factor for primary graft nonfunction.Conclusion: This survey highlights the significant variation in the appraisal and use of steatotic grafts by LT surgeons in ANZ. Accurate evaluation and judicious use of mild and moderately steatotic grafts is required if we are to utilize the available donor pool best.Keywords: liver transplantation, steatosis, fatty liver, organ donors. Canada balsam is secreted by cells in the bark of the balsam fir Abies balsamea, a conifer widely distributed across Canada and northern U.S.A. Collected by an extraordinarily labour-intensive process, it was initially exported to Europe for use in pharmacy. However, it found a much wider application after Brewster showed that the refractive index of 1·54–1·55 characterizing the vitreous mass remaining after heating the natural product exceeded that of most resins, and therefore closely matched the indices of ‘crown’ and (...) ‘light flint’ optical glasses. ‘Cooked’ Canada balsam was therefore very suitable for cementing achromatic lenses for telescopes and microscopes, as well as for mounting specimens for microscopical examination. Canada balsam remained pre-eminent for over a century, but the rise of synthetic adhesives and other substitutes has now relegated the natural balsam to a few specialized niches. (shrink)

Concept of Avidyā in the Vedānta philosophy of Śaṅkarācārya; a study.

The most prominent form of human communication and interaction is speech. It plays an indispensable role for expressing emotions, motivating, guiding, and cheering. An ill-intentioned speech can mislead people, societies, and even a nation. A misguided speech can trigger social controversy and can result in violent activities. Every day, there are a lot of speeches being delivered around the world, which are quite impractical to inspect manually. In order to prevent any vicious action resulting from any misguided speech, the development (...) of an automatic system that can efficiently detect suspicious speech has become imperative. In this study, we have presented a framework for acquisition of speech along with the location of the speaker, converting the speeches into texts and, finally, we have proposed a system based on long short-term memory which is a variant of recurrent neural network to classify speeches into suspicious and nonsuspicious. We have considered speeches of Bangla language and developed our own dataset that contains about 5000 suspicious and nonsuspicious samples for training and validating our model. A comparative analysis of accuracy among other machine learning algorithms such as logistic regression, SVM, KNN, Naive Bayes, and decision tree is performed in order to evaluate the effectiveness of the system. The experimental results show that our proposed deep learning-based model provides the highest accuracy compared to other algorithms. (shrink)

This study discusses the relationship between Green Chemistry and Environmental Sustainability as expressed in textbooks and articles on Green Chemistry authored by their promoters. It was found that although the Brundtland concept of Sustainable Development/Sustainability has been mentioned often by green chemists, a full analysis of that relationship was almost never attempted. In particular, green chemists have paid scarce attention to the importance of The Second Law of thermodynamics on Environmental Sustainability and the consequences of the limitations it imposes on (...) Background: One of the barriers to interprofessional ethics education is a lack of resources that actively engage students in reflection on living an ethical professional life. This project implemented and evaluated an innovative resource for interprofessional ethics education. Objectives: The objective of this project was to create and evaluate an interprofessional learning activity on professionalism, clinical ethics, and research ethics. Design: The Brewsters is a choose-your-own-adventure novel that addresses professionalism, clinical ethics, and research ethics. For the pilot of the book, (...) a pre-test/post-test design was used. Once implemented across campus, a post-test was used to evaluate student learning in addition to a student satisfaction survey. Participants and research context: A total of 755 students in six academic schools in a health science center completed the activity as part of orientation or in coursework. Ethical considerations: The project was approved as exempt by the university’s Committee for the Protection of Human Subjects. Findings: The pilot study with 112 students demonstrated a significant increase in student knowledge. The 755 students who participated in the project had relatively high knowledge scores on the post-test and evaluated the activity positively. Discussion: Students who read The Brewsters scored well on the post-test and had the highest scores on clinical ethics. Clinical ethics scores may indicate issues encountered in mass media. Conclusion: The Brewsters is an innovative resource for teaching interprofessional ethics and professionalism. Further work is needed to determine whether actual and long-term behavior is affected by the activity. (shrink)

This original and important work is a sequel to Conceptual Idealism and The Primacy of Practice. It is a systematic treatment of the "problem of the legitimation of factual knowledge"-a central issue of rational warrant or cognitive rationality. It offers a doctrine of methodological pragmatism, as distinct from theses pragmatism—a distinction analogous to act- and rule-utilitarianism. Theses pragmatism "asserts that a proposition is to be accepted if its adoption is maximally success-promoting," whereas methodological pragmatism "asserts that a proposition is to (...) be accepted if it conforms to an epistemologically warranted criterion and that a criterion is warranted if its adoption as a generic principle for propositional acceptance is maximally success-promoting ". Thus what is offered is a pragmatic justification of cognitive methodology in factual inquiry rather than the pragmatic validation of factual theses. (shrink)
